You probably need to know the diameter or circumference of the disc.
For example, if the disc has a diameter of 2 cm, then the radius is 1 cm = 10000 micrometers.
That means the area of the disc is pi * 10000^2.
Since you said the thickness is 100 micrometers, the volume of the disc would then be 100 * pi * 10000^2 = 1e10 * pi cubic micrometers.
Now just convert cubic micrometers to microlitres:

1e10 * pi cubic micrometers = 10 * pi microlitres
So for a disc with a radius of 1 centimeter, the volume is 10 * pi microlitres.
You'll have to adjust this calculation if you are working with a different size disc.
